    Function annotation of hepatic retinoid x receptor α based on genome-wide DNA binding and transcriptome profiling.

    BackgroundRetinoid x receptor α (RXRα) is abundantly expressed in the liver and is essential for the function of other nuclear receptors. Using chromatin immunoprecipitation sequencing and mRNA profiling data generated from wild type and RXRα-null mouse livers, the current study identifies the bona-fide hepatic RXRα targets and biological pathways. In addition, based on binding and motif analysis, the molecular mechanism by which RXRα regulates hepatic genes is elucidated in a high-throughput manner.Principal findingsClose to 80% of hepatic expressed genes were bound by RXRα, while 16% were expressed in an RXRα-dependent manner. Motif analysis predicted direct repeat with a spacer of one nucleotide as the most prevalent RXRα binding site. Many of the 500 strongest binding motifs overlapped with the binding motif of specific protein 1. Biological functional analysis of RXRα-dependent genes revealed that hepatic RXRα deficiency mainly resulted in up-regulation of steroid and cholesterol biosynthesis-related genes and down-regulation of translation- as well as anti-apoptosis-related genes. Furthermore, RXRα bound to many genes that encode nuclear receptors and their cofactors suggesting the central role of RXRα in regulating nuclear receptor-mediated pathways.ConclusionsThis study establishes the relationship between RXRα DNA binding and hepatic gene expression. RXRα binds extensively to the mouse genome. However, DNA binding does not necessarily affect the basal mRNA level. In addition to metabolism, RXRα dictates the expression of genes that regulate RNA processing, translation, and protein folding illustrating the novel roles of hepatic RXRα in post-transcriptional regulation

    Surface anesthesia of the small incision cataract extraction and intraocular lens implantation

    AIM: To observe the effect of cataract extraction and intraocular lens implantation with manual small incision cataract surgery under surface anesthesia in the Primary Hospitals.<p>METHODS: Totally, 372 eyes of 358 cataract cases, in our hospital from January 2010 to March 2013, underwent cataract extraction and intraocular lens implantation with manual small incision cataract surgery under surface anesthesia. Intraoperative pain, postoperative complications and visual acuity were observed.<p>RESULTS: In the process of surgery, patients without pain and being-cooperated were recorded in 342 eyes(91.9%), that occasionally felt slight pain, but within endure and the operation was completed successfully were recorded in 17 eyes(4.6%). There were 13 eyes quit, 8 eyes(2.2%)of them because of the intolerable pain, the other 5 eyes(1.3%)of the ceaseless movement of the eye. The best corrected visual acuity on the 3<sup>rd</sup> day after operation of 4.0~4.5 were observed in 57 eyes(15.9%), 4.5~4.8 in 213 eyes(59.3%), above 4.8 in 89 eyes(24.8%).<p>CONCLUSION: The cataract extraction and intraocular lens implantation with manual small incision cataract surgery under surface anesthesia can avoid corresponding complications, reduce the operation time obviously and ensure better quality of operation. It's suitable to carry out the prevention and treatment of blindness at the Primary Hospitals

    Exploring Online Double Deviation Effect from Psychological Contract Violation, Emotion, and Power Perspectives

    Service recovery is a critical moment of truth in retaining customers and reinforcing customer relationships, and has been considered as an “Achilles\u27 heel” in online marketplaces. Poor service recoveries exacerbate the negative effects of the failure, producing a “double deviation” effect. The double deviation effect may arise from the seller’s power misuse and then dissolve the buyer-seller relationship (e.g., violate consumer psychological contract), elicit consumer negative emotions which lead to customer coping behaviors. This study links the theories of psychological contract violation (PCV), emotion, and coping from the power perspective to investigate the double deviation scenario in online auction marketplaces. Two moderators (perceived power and perceived consumer empowerment) are considered in our proposed model. Data collected from 190 consumers of one auction website provide support for the proposed model. The results shed light on what constitutes the determinants of consumer judgments while facing double deviation scenario and how consumers react to and cope with it in online marketplaces.     Anomalous pressure behavior of tangential modes in single-wall carbon nanotubes

    Using the molecular dynamics simulations and the force constant model we have studied the Raman-active tangential modes (TMs) of a (10, 0) single-wall carbon nanotube (SWNT) under hydrostatic pressure. With increasing pressure, the atomic motions in the three TMs present obvious diversities. The pressure derivative of E1g, A1g, and E2g mode frequency shows an increased value (), a constant value (), and a negative value () above 5.3 GPa, respectively. The intrinsic characteristics of TMs consumedly help to understand the essence of the experimental T band of CNT. The anomalous pressure behavior of the TMs frequencies may be originated from the tube symmetry alteration from D10h to D2h then to C2h.Comment: 15 pages, 3 pages, submitted to Phys. Rev.
